Display-based antibody discovery & engineering

We pair surface-display platforms with FACS-based selection to mine antigen-specific antibodies from large libraries and to engineer them for higher affinity, broader reactivity, and better developability — turning repertoire insight into therapeutic candidates.

Yeast surface display platform for antibody discovery and engineering. Paired antibody VH:VL Fabs are expressed as Aga2 fusions displayed on the yeast surface with FLAG and c-Myc tags; iterative FACS sorting against fluorescent antigen (shown here for influenza hemagglutinin) enriches antigen-specific binders across successive rounds, yielding engineered antibodies characterized for IgG affinity and neutralization potency.

Discovery — mining rare binders

Large synthetic and immune libraries are displayed and screened by FACS against fluorescent antigen, recovering rare, high-affinity, antigen-specific clones that conventional screening often misses.

Engineering — maturing function

Iterative rounds of mutagenesis and selection mature antibodies for affinity, cross-reactive breadth, and biophysical developability, advancing them toward diagnostic and therapeutic candidates.
